Somatic mutations in genes encoding RNA splicing factors are prevalent across various hematologic malignancies, including myelodysplastic syndromes and acute myeloid leukemia. Perturbed RNA splicing is often associated with an abnormal accumulation of RNA:DNA hybrid structures known as R-loops, depicted in the bottom left of the cover image. Spliceosome-mutant leukemias are dependent on PARP1 for survival due to its role in suppressing transcription–replication conflicts at R-loops, highlighting the potential therapeutic utility of PARP inhibitors in the treatment of hematologic malignancies.
